Synthesis and anti-tumor activity of 5 fluoro-2-Methy1-6(4-(substituted pheny1) piperazin-1-y1) benzoxazoles

A novel series of 5-fluoro-2-methyl-6-(4-(substituted phenyl)piperazin-1-yl) "benzoxazoles were prepared. The synthetic strategy includes three steps. Controlled nitration of 3-chloro-4-fluorophenol followed by piperazinylation yielded a new series of intermediates, the 4-fluoro-5-(4-substituted phenylpiperazin-1-yl)-2nitrophenols. The target benzoxazoles were synthesized through reduction of the latter using indium and acetic acid followed by cyclization of 4-fluoro-5-(4substituted phenylpiperazin-1-yl)-2-aminophenols with trimethyl orthoacetate. The structures of all synthesized final products and intermediates were confirmed using IR, 'H and '3C NMR and HRMS spectral analysis. The preliminary biological results of intermediates showed promising anticancer activities.
